Our group is interested in understanding the molecular architecture of proteins using biophysical methods, molecular dynamics simulations, molecular modeling and crystallographic approaches. The knowledge of protein structure is further utilized for the design of potential drug like compounds for the treatment of life threatening diseases.

Structure Determination of Drug Targets

Structure-based design is one of the best approach used in drug design. We used X-ray crystallography to determine the structure of potential drug targets which are employed in the discovery process of new drugs. The protein sequence→structure→function relationship is well established and reveals that the structural details at atomic level help understand molecular function of proteins.

Search for Potential Inhibitors using structure based approach
Different methods of computer-aided drug discovery are used to find novel inhibitors for the target protein. We employee molecular docking, high throughput screening, virtual screening and QSAR to search potential inhibitors of a particular drug targets (specifically human kinases).

Experimental validation of Ligand-Receptor interactions
Protein of interest (drug target) is being cloned and expressed in our lab. We use fluorescence and ITC methods to determine the binding affinity of selected ligands to the particular receptors. We further checks the efficacy of selected inhibitors at cellular level.
Functional knockout of target genes
The knockout of protein function for individual genes (drug target) is a powerful tool to further validate the impact of inhibitors on the particular biological process. CRISPR-based methods are implemented to knock out the gene function, specific edits to the genome, and activation or inhibition of gene expression.
